The ~以上は and ~上は patterns are both used to express "as long as/since/seeing as...", and are fairly stiff in tone.
~以上・上は statements tend to express things like "since X, it's obvious that Y" or "since X, (I) will certainly do Y", or similar.
E.g. 証拠が見つかった以上、もう言い逃れはできないだろう。 "Seeing as how they have found evidence, they shouldn't be able to get away now."
E.g. 人数が少ない以上、役割はきちんと決めるべきです。 "As long as there are few members, (we) ought to carefully decide roles."
E.g. キャプテンである以上、部員のロールモデルでありたい。 "As long as I'm the captain, I want to be a role model for the club members."
~上は is more stiff/formal than ~以上は, which is more polite/formal in tone.
E.g. 契約を締結した上は、必ず遂行せねばならない。 "Now that we have entered into a contract, I must execute no matter what."
は may be dropped from ~以上は and ~上は, most commonly in written scenarios.
~以上は and ~上は attach directly to casual form verbs/い-adjectives, or to nouns/な-adjectives using である.
